Introducción: Las hendiduras orofaciales, como el labio y paladar fisurado, son malformaciones congénitas más comunes, con una alta incidencia y prevalencia a nivel mundial. El tratamiento de las HO, inicia inmediatamente después del nacimiento, hasta la adolescencia, incluso hasta la edad adulta, mediante múltiples intervenciones pre-quirúrgicas, quirúrgicas y postquirúrgicas. Objetivos: El objetivo de este estudio fue evaluar mediante una revisión de literatura la etiología, diagnóstico y tratamiento de labio, paladar y labio/paladar fisurado en neonatos y lactantes. Materiales y Métodos: La literatura se seleccionó mediante una búsqueda en las bases de datos electrónicas: ScienceDirect, PubMed, SciELO, Google Scholar, y los informes de la Organización Mundial de la Salud (OMS), plataformas de American Chef Palate (ACPA) - Craniofacial Association Centers for Disease Control and Prevention, WebMD, y el portal de Información de Enfermedades Raras y Medicamentos Huérfanos. La búsqueda se restringió a artículos en inglés y español, publicados desde el año 2011 al 2021. Resultados: Después de aplicar los criterios de inclusión, en total se obtuvieron y revisaron 35 artículos. Se realizó la revisión de literatura de la etiología, diagnóstico y tratamiento del labio, paladar y labio/paladar fisurado en neonatos y lactantes. Conclusiones: Los autores consideran que la etiología principal del labio y paladar fisurado es desconocida; sin embargo, existen diversos factores de riesgo genéticos y no genéticos. El diagnóstico se realiza mediante la exploración ecográfica a las 20 semanas de vida intra-uterina, y es indispensable la coordinación de los equipos multidisciplinarios para garantizar los mejores resultados.
